Excerpt:<br>Brucellosis exomembrane protein OMP25 is an important immunogenandamy and protective antigen in brucellosis, which can prevent the production of pro-inflammatory cytokines, promote the survival of the bacteria and immune escape. The OMP25 protein is conservatively present in all major brucellosis and is one of the promising subunit vaccines. In this study, the olyscocci OMP25 gene from clinical patient identification culture was cloned and protein expression, and the OMP25 complete gene was successfully obtained, coding the base because 657bp, Sequencing analysis showed that the OMP25 gene sequence of brucellosis encoded in the NCBI data bank was 100% homogenous, the expression product signed by SDS-PAGE and Western blot analysis, which proved to be consistent with theoretical calculations, was about 24.1 kDaa, and the Expression yield of OMP25 protein in E.Coli was up to 200mg/L. The advanced structure of OMP25 protein is predicted by software, and the antigen epitope of OMP25 protein is predicted and analyzed. The research results lay the foundation for the preparation of OMP25 subunit vaccine.
