Abstract: <br>Brucella outer membrane protein OMP25 is an important Brucella immunogenicity and protective antigen, it can prevent the production of pro-inflammatory cytokines, promote cell survival and immune evasion. OMP25 are conserved protein found in all the major Brucella, it is one of the candidate subunit vaccine have developed promising. In this study Brucella OMP25 genes from patients with clinical training were identified and cloned protein expression, successfully acquired a complete OMP25 gene, encoding gene is 657bp, sequencing analysis showed that Brucella coding and published NCBI Repository OMP25 gene homology of 100%; the expressed product by SDS-PAGE and Western blot analysis proved consistent with the theoretical value of about 24.1 kDa; Ni-NTA column purification and anti purified by, the OMP25 protein in E.Coli expression yield of up to 200mg / L. Through advanced software to predict the structure of OMP25 protein epitope prediction analysis of OMP25 protein. This research laid the foundation for the preparation of OMP25 subunit vaccine.
